The uncontrolled activation of kinases and the suppression of phosphatases has been frequently observed in cancer with consequent induction of cell proliferation, migration and survival to anti-cancer therapies. Moreover, oncogenes can influence the balance between kinases and phosphatases activity, causing cell malignant transformation ( Qi et al., 2018). Mutations in kinases ( Irby et al., 1999 Zhang et al., 2009) or phosphatases ( Stebbing et al., 2014 Zhao et al., 2015) that lead to either a loss-of-function or gain-of-function are likely to cause cancer. The main role of kinases and phosphatases is to regulate post-translational modifications of proteins, which are essential to govern cellular signaling networks ( Sacco et al., 2012). Moreover, we discuss the potential of inhibitors of these proteins in counteracting CSCs expansion during cancer development and progression. Here, we provide a comprehensive overview of the major protein kinases and phosphatases pathways by which CSCs can evade normal physiological constraints on survival, growth, and invasion. Notwithstanding the role of kinases and phosphatases in cancer has been widely investigated, their aberrant activation in the compartment of CSCs is nowadays being explored as new potential Achille’s heel to strike. Therefore, alterations in kinases and phosphatases functionality is a hallmark of cancer. Fret not though parents, because surprisingly this film will have adult moments placed in as well, comedy lines and items that you'll quite enjoy at the fourth-dimension breaking or genre poking moments that are very funny. ![]() Slapstick humor and annoying lines are in this movie, and as my showing showed are very repeatable by those who like to parrot lines. The trailers show it's primarily for kids, and to be honest, it does have a lot of things geared towards a younger audience. The Humor: Now we are getting to my favorite elements of the bunch, the humor of the movie. All in all, this cast did better than I thought and could play very well together for a future, most likely impending series. I can't say he did the best of the bunch, but his style of working with all the other voice actors was very balanced, and always maintained that comedic style I love to see. He does Krypto decently, giving the kid humor element the biggest punch of the actors, while sometimes nailing those overdramatic moments that a superhero film is noted to have. As for Johnson, he's still the same guy you love to watch on the screen, only in a CGI, white dog form. He does well to play with the Rock as expected, and this movie keeps their friendship in real life well-integrated into the characters of Krypto and Ace to the point of being very entertaining. He rants, does montages with the same type of fire, and his comedy about being put into situations was very fun, but I loved seeing the vulnerable side come out with it to help balance the comedy and character of Ace. Hart is also pretty much playing his deck of cards again that he has made him so famous. It works well for this type of movie and I had fun hearing her play the sassy self in animal form quite well. If you love seeing, or in this case, hearing her rants and overenthusiastic delivery, get ready for about 90 minutes of it. McKinnon is a master of so many roles, and Lulu is no exception as the hairless guinea pig role that is like one of her SNL skits come to life, only censored. Yet the big three get the focus, so let's center on them for a moment. While Natasha Lyonne may have been my favorite performance of the bunch with her balanced humor and turtle like mafia voice. John Krasinski as Superman was sensational in his fun and yet mature manner that was like serious Jim from the Office. Voice acting is an amazing art as I've said before, and so many people accomplished the directions they were taking with various parties in the movie. The Voice Acting: Alright now onto the people who brought the character part of the group to life. The movie has plenty of family friendly moments that can melt many hearts, and will probably inspire you to be kind to your pets or adopt. Cursing/semi-swear words are at a minimum, dark natured elements are toned down to G or PG levels, and there should be very few things to scare or scar your kids. The Cute Factor: I could go on for hours about the cute factor of this movie, but parents rest in relief, because the movie is indeed very cuddly for your group. ![]() All of it works together to make the adventure fun and enjoyable to watch, without feeling stuck in an outdated computer game. While there is definitely some realism to it though, the designers did keep the comic book/cartoon mojo to it and I was impressed with how much of a little kid I felt in me while watching it. It's smooth, it does not take too many clunky shortcuts to get the shot, and works well to match the style of the character and voice actor. DC Super-pets has a nice flow that allows there characters to move in the heroic style you expect. I use an Akai IOdock with my iPad and did not drag my iPod Touches along for thae show. The sounds I created worked well and we got a tremendous response from an audience of 35 or so (it was a very small venue/ very large living room). I recently used my iPad at a gig I played with The Dits a free improv/noisy/trippy/humorous trio I have played with for the last 12 years except for a not so brief hiatus for back surgery. I also use some sound maker type apps like Slewpi, Syntpond, Reactable, Keplers orrey, Slice, Aura Flux and iGendyn for improvisation and inspiration. Moog has 2 apps I use a lot called Filtatron and AniMoog which create sounds worthy of the Moog name. 1 cent accuracy as their hardware brethren ideal for tuning or intonation adjustment. ![]() My main tuner is an app by Peterson called iStrobsoft that gives the same. Another cool one is Droneo which creates drones that can be tweaked to the nth degree. There are also a host of strange synthy type apps from Curtis that I use a lot that have unique interfaces and can bend sounds dramatically. I also use an app called Teslas Synth which is similar but a bit more aggressive. It is 4-5 note polyphonic and looks like a kids toy but it is very expressive. ![]() Some of my faves are Bebot which is a synth whose interface is a stupid looking little robot that moves as you place your finger on the screen left for low note right for higher notes as you move up the modulation increases you can have either continuous note change or add lines for a more keyboard like response. Parallels obviously taxes the battery life, so I do plenty of stuff outside Parallels, and then hop to Windows for specific needs: PowerShell, C# development, specific Windows software such as Power BI Desktop and similar. ![]() Why not just use Windows 11 all day long? Parallels 19 brings support for SQL Server and x86 Docker containers - so obviously if your work includes running stuff like this locally, you might need to allocate more RAM for the VM. Then again, I rarely need a VM with 32 or 64 GB of RAM - and of I do, it's typically not something I do a on laptop, but just push those workloads in the cloud. If I know I'll be doing more software development, I opt for maybe 16 GB on the Windows 11 instance. I'm currently running a single Windows 11 VM with 8 GB of RAM. In the image above, you can see that I've used about 24 GB of RAM. Parallels is pretty aggressive with default settings in suspending VMs that are idle, but even if I disable that - nothing tells me that the MacBook Pro is running another OS on the side. This is weird, but there isn't a way for me to verify if I have a VM running, unless I go and see specifically if it's running. Will macOS slow down when a Windows 11 VM is running? Once in full-screen, it's identical to using Windows 11 on a regular PC. This approach allows me to quickly swipe to that desktop, or I can swipe three fingers up, and just hop to a specific desktop. I usually dedicate the last virtual desktop for it, so if I have 3 virtual desktops already in macOS, the Parallels-based VM will be the fourth virtual desktop. Once you have a VM running, you can dedicate a virtual desktop for it, or run it in a window as a regular app. It's super convenient - almost like Windows Hello on a Surface device. How it works is that if you set a Windows Hello PIN, when you're logging in to your VM, you can just swipe the Touch ID button instead of typing in a PIN.
